In this lab, students perform DNA electrophoresis and examine gels for a mother, baby and possible fathers to determine correct paternity. Kit contains gel boxes, power supplies, dry heat bath, micropipettes, latex gloves, dark reader, and all other supplies necessary to complete the included lab. A separate Pipette Practice Kit, and extra supplies for large classes, are available upon request.
Students will cut DNA with restriction enzymes. The DNA fragments will be separated electrophoretically on an agarose gel. And finally, the results will simulate a DNA profile.
KIT DETAILS
The kit includes:
- 4 gel boxes with trays, combs and wires
- 2 power supplies
- 8 micropipettors (2-20 μl)
- 1 micropipettor (20-200 μl)
- 1 dry bath for 1.5 mL Eppendorf® tubes, with block
- 1 thermometer
- 8 Eppendorf® tube racks
- 1 centrifuge
- 4 boxes micropipette tips
- 2 bags 1.5 mL Eppendorf® tubes (at least 100 tubes)
- 1 fridge pack with expendables
- 1 bottle (20 g) agarose
- 1 bottle TBE running buffer
- 1 dark reader
- 1 fuse
- Kit notebook
- CD with a PowerPoint DNA presentation

DNA Profiling Reagents (per 16 students):
- Sterile H2O (1000 µl)
- Buffer (150 µl)
- M3 (50 µl)
- S3 (50 µl)
- B3 (50 µl)
- M4 (50 µl)
- S4 (50 µl)
- B4 (50 µl)
- HindIII (45 µl)
- Loading Dye (400 µl)
- HindIII lambda DNA (40 µl)
- GelStar DNA Stain (12 µl)
Teachers must supply:
- Hot plate with magnetic stirrer (or a microwave oven)
- Two 2 L flasks and 100 mL graduated cylinders
- Four 250 mL flasks
- 4 L distilled H2O
- 1 box latex gloves
- Waste containers
- Sharpies, various colors
- Ice or a nearby freezer
- Semi-log paper or access to a computer graphing program
